Regulation of androgen receptor in male development.

 

A major focus of our work is to uncover the mechanism by which the androgen steroid hormone receptor directs gene transcription in response to cellular context. The androgen receptor (AR) directs many aspects of human biology including male development and prostate physiology. The AR acts in conjunction with chromatin binding proteins that regulate polymerase action and recruitment of AR to target genes. We have recently characterized a new repression complex that contains Unconventional prefoldin RPB5 Interactor (URI), a component of the androgen receptor transcription complex modified in response to hormones, DNA damage and nutrient uptake. Using mouse models, we have shown that URI and interacting binding proteins are important for cell lineage specification and critical for testicular germ cell maturation. We are also collaborating with the DasGupta laboratory to understand the synergistic role of beta-catenin and AR in prostate cancer and cancer initiating cells.
